WHAT IS IT?
Anamorelin is a non-peptide, orally-active, centrally-penetrant, selective agonist of the ghrelin/growth hormone secretagogue receptor (GHSR) with appetite-enhancing and anabolic effects.
Ibutamoren is a potent, long-acting, orally-active, selective, and non-peptide agonist of the ghrelin receptor and a growth hormone secretagogue, mimicking the growth hormone (GH)-stimulating action of the endogenous hormone ghrelin.
HOW DOES IT WORK?
Anamorelin significantly increases plasma levels of growth hormone (GH), insulin-like growth factor 1 (IGF-1), and insulin-like growth factor-binding protein 3 (IGFBP-3) in humans, without affecting plasma levels of prolactin, cortisol, insulin, glucose, adrenocorticotropic hormone (ACTH), luteinizing hormone (LH), follicle-stimulating hormone (FSH), or thyroid-stimulating hormone (TSH). In addition, anamorelin significantly increases appetite, overall body weight, lean body mass, and muscle strength, with increases in body weight correlating directly with increases in plasma IGF-1 levels.
Ibutamoren (MK-677) has been shown to sustain activation of GH–IGF-1 and to increase lean body mass with no change in total fat mass or visceral fat. It is under investigation as a potential treatment for reduced levels of these hormones. Human studies have shown ibutamoren to increase both muscle mass and bone mineral density, making it a promising potential therapy for the treatment of frailty in the elderly.
Growth Hormone acts both directly and by stimulating IGF-1. When released, GH travels from the anterior pituitary gland to liver and cartilage via bloodstream and bind to its receptor, Growth Hormone Receptor (GHR), which triggers production of Insulin-Like Factor 1 (IGF-1).
Both GH and IGF-1 are needed to stimulate normal linear growth. Growth Hormone acts at the growth plate and increases linear growth by promoting differentiation and expansion of cells. It increases bone mass by stimulating proliferation of bone cells, causing bone accumulation. GH acts on the fat tissue to increase the breakdown of fat and mobilization. It also increases transport of amino acids in muscles to affect muscle fiber distribution.
