Description

Sildenafil

PDE-5 Inhibitor Research Compound

Sildenafil is a selective phosphodiesterase type 5 (PDE-5) inhibitor extensively studied for its effects on nitric oxide signaling, cyclic guanosine monophosphate (cGMP), vascular smooth-muscle relaxation, and blood flow.

Compared with longer-acting PDE-5 inhibitors such as tadalafil, sildenafil has a shorter duration of activity, making it useful in research examining more acute changes in PDE-5 inhibition and vascular physiology.

How Sildenafil Works

Sildenafil inhibits the PDE-5 enzyme, which is responsible for breaking down cGMP. By slowing cGMP degradation, sildenafil can enhance nitric oxide–mediated signaling and promote relaxation of vascular smooth muscle.

This mechanism has made sildenafil an important compound in research involving:

  • PDE-5 inhibition
  • Nitric oxide and cGMP signaling
  • Vascular smooth-muscle physiology
  • Blood-flow regulation
  • Endothelial function
  • Pulmonary vascular physiology

Key Characteristics

Potent PDE-5 Inhibition
Sildenafil selectively inhibits PDE-5 and has become one of the most extensively characterized compounds within this pharmacological class.

Shorter Half-Life
Sildenafil has an elimination half-life of approximately 4 hours, providing a substantially shorter activity profile than tadalafil.

Nitric Oxide Pathway Research
Its effects on the nitric oxide–cGMP pathway make sildenafil valuable for studying vascular signaling and smooth-muscle responses.

Extensively Studied
Sildenafil has decades of clinical and pharmacological research behind it, providing a substantial body of scientific literature regarding its pharmacokinetics, mechanisms, and physiological effects.

Research Areas

Sildenafil has been investigated in research related to:

  • Nitric oxide signaling
  • cGMP metabolism
  • PDE-5 enzyme activity
  • Vascular and endothelial function
  • Smooth-muscle physiology
  • Pulmonary circulation
  • Blood-flow regulation
  • Exercise physiology
  • Erectile physiology
  • Cardiovascular signaling pathways

Compound Information

Compound: Sildenafil
Compound Class: PDE-5 Inhibitor
Molecular Formula: C₂₂H₃₀N₆O₄S
Molecular Weight: 474.6 g/mol
Approximate Half-Life: 3–5 hours
Primary Target: Phosphodiesterase Type 5 (PDE-5)

Sildenafil vs. Tadalafil

While both compounds inhibit PDE-5, one of the primary differences is their pharmacokinetic profile. Sildenafil has an approximate half-life of 3–5 hours, while tadalafil has an approximate half-life of 17.5 hours. This makes sildenafil particularly relevant for research examining shorter-duration PDE-5 inhibition.
